The ideal grain used for equatorial orientation should have the aperture (if present) at the highest or first plane of focus. Grains selected for each of these views should be direct orientations or rotated as little as possible. In general, two Z-Stacks of each species should be obtained: one from the equatorial orientation and one from the polar orientation. This protocol is used to capture Z-Stack images of pollen grains.
